The spare tire is smaller diameter, so your engine is at slightly higher RPM (if its on the front) to get you to speed. Also, the temp tire is made of a very soft and cheap rubber, so there is high rolling resistance.

yes it will affect your gas mileage, greatly so, that is why they are only suppose to be driven on a max of 50 miles. Just enough to get you to the tire shop to get it replaced!

Yes your car was tested with the stock parts a spread is a different sizes and will affect MPG just as carrying a heavy load will or upsizing the rims

That much lessening in MPG seems quite excessive. Remount your running wheel and tire and recheck. It seems possible another fault causing that fuel mileage drop, but you should clear the faulty (spare tire on) condition to find out for sure.

Yes, you can certainly expect a drop in fuel efficiency. More importantly, if you are using a compact spare it is only meant to be used very temporarily. 50 miles or so, max, and at a top speed of 50 mph. Compact spares are simply not designed to be pushed beyond these limits. Replace it asap.
