How do you think they can run DNA tests and figure out if people are related? Blood tests (among other DNA tests such as using spit) links people together who are kin.

You should really ask this in the biology section, genealogy is about tracing your family tree. My opinion - for what it's worth - is that the whole blood thing is just another way of saying you are related. A child can have a different blood group to it's parent so of course the blood isn't exactly the same. However a child does have half of each parent's genes. Maybe in the story twice as much blood must be used because it only has half the magic property?

No, it's a metaphor. Even in the uterus, a fetus' blood does not mix with its mother's blood. But similarity in DNA due to common ancestors is often referred to as "consanguinity" (sharing blood). But, again, that's a metaphor. Often, members of the same biological nuclear family even have different blood types, so that one can't give a blood transfusion to the other.

Not exactly Direct line relatives share the same "blood-line" and genetic patterns. Your story has nothing to do with Genealogy and since it is a fiction - anything is possible that is what fiction is about. It is make believe and not about plausibility.

It is not plausible, could never be plausible, but would work in a fantasy-type story. No, people are not related by blood; in olden times, people thought that the heart was the brain/mind; they did not know how a woman got pregnant, did not know about germs, or about DNA. You are related to any relative through DNA, not blood.
